pir gene
The pir gene encodes the π protein. Initiation of replication at ori-γ or other R6K origins of replication requires the π protein in trans. Increasing its expression to double the natural concentration produced would, however, effectively shut down the replication. Expression of π protein is autogenously regulated.
Usage and Biology
The presence of pir gene allow maintenance of plasmids with R6K origins as their sole origin of replication.
